摘要
目的研究负载表柔比星的磁性壳聚糖纳米粒(CS-PAA-EPI)的全身分布及器官靶向性。方法选用壳聚糖、磁流体及表柔比星为原材料首次合成CS-PAA-EPI,运用高效液相法(HPLC)检测CSPAA-EPI在大鼠体内各主要脏器的药物浓度,进而评价CS-PAA-EPI的器官靶向性。结果与对照组相比,在外加磁场的作用下,实验组大鼠EPI在给药后5、15、25、30、40、60、180min的膀胱靶向率为:30%、55.4%、76.4%、74.3%、69.2%、56.6%、45.2%,提示给药后5min就明显的聚集到膀胱内,在给药后25min后其靶向分布率最高为76.4%,各个时间点实验组大鼠膀胱EPI靶向率均明显高于对照组(P<0.01)。结论在外加磁场的作用下,磁性壳聚糖纳米粒可以很好的将表柔比星主动靶向于膀胱壁全层,从而明显减少了全身重要脏器内的分布,尤其是肝、脾的被动靶向,在不同时间点,实验组大鼠膀胱的靶向率均明显高于对照组大鼠,为后续的研究提供理论支持。
Objective To investigate determination CS-PAA-EPI Polymer Magnetic Nanopaticle in mouse plasma.Methods Magnetic CS-PAA-EPI Nanopaticle was composed by CS,acrylic monomer(AA),EPI and Fe_3O_4 nanopaticle.HPLC was used to investigate determination of CS-PAA-EPI polymer magnetic nannopaticle in mouse plasma.Results To compare with control group,the EPI targeted rate in mouse bladder in experimental group(30%,55.4%,76.4%,74.3%,69.2%,56.6%,45.2%,5 min,15 min,25 min,30 min,40 min,60 min,180 min after drug injected)were significantly increased.Conclusions With the reaction of magnetic field, CS-PAA-EPI Polymer Magnetic Nanopaticle can concentrate in mouse bladder,which supply great support for drug target chemotherapy.
