NY-ESO-1基因在膀胱癌中的表达及临床意义的研究

目的探讨NY-ES0-1的表达与膀胱癌分期分级的关系以及临床意义。方法回顾性研究2001～2002年膀胱癌组织32例,其中男性24例,女性8例。正常膀胱粘膜对照取自耻骨上经膀胱前列腺切除术的良性前列腺增生的患者。阳性对照为正常睾丸组织。常规行RT-PCR方法检测膀胱癌组织及对照组等NY-ESO-1基因的表达,分析其表达与膀胱癌分期分级的关系以及临床意义。结果 NY-ESO-1 mRNA在32例膀胱癌组织中12例表达(37.5%),在正常膀胱粘膜上没有表达。肿瘤组织中PCR反应的产物与正常睾丸的表达产物一致。NY-ES0-1 mRNA的表达和膀胱癌的病理分级相关,有统计学差异(P<0.05)。NY-ESO-1 mRNA的表达率随着肿瘤病理分级的增高而升高:G1期是0/3(0%);G2期是4/16(25%);G3期是8/13(61.54%)。浸润性膀胱癌中NY-ESO-1 mRNA的表达与表浅性肿瘤的表达无显著性差异。结论 NY-ESO-1 mRNA在正常膀胱组织上没有表达,仅在正常睾丸组织和病理膀胱癌组织上有表达。NY-ESO-1 mRNA的表达率随着膀胱癌病理分级的增高而升高。

Objective To evaluate NY-ESO-1 mRNA expression in both the patients of bladder Transitional-cell cancer(TCC)and the healthy volunteers and to explore the correlation between NY-ESO-1 expression and tumor stages and grades.Methods In this study,bladder cancer specimens were obtained from 32 patients undergoing surgery,consisting of 24 males and 8 females.Reverse-transcription(RT)-PCR was performed to evaluate NY-ESO-1 mRNA expression in the tissues of bladder TCC,testis tissue and the healthy volunteers.The correlation between NY-ESO-1 gene expression and tumor stages and grades were explored.Results NY-ESO-1 mRNA was detected in 12 of 32(37.5%)tumor specimens without its expression in the normal bladder tissues.Higher frequency of NY-ESO-1 mRNA expression was seen in higher grade TCC;0 of 3(0%)in G1;4 of 16(25%)in G2;8 of 13(61.54%).No correlation could be established between NY-ESO-1 mRNA expression and the presence or absence of muscle invasion. Conclusions NY-ESO-1 gene does not express in normal bladder tissue,but express in both bladder transitional cell cancer and normal testis tissue.Bladder TCC is a tumor type with a high frequency of NY-ESO -1 mRNA expression.Higher frequency of NY-ESO-1 mRNA expression was seen in higher grade TCC. There is no difference of NY-ESO-1 mRNA expression between the superficial and invasive bladder cancer.
