摘要
To investigate the role of basic fibroblast growth factor(bFGF) in inducing the differentiation of mesenchymal stem cells (MSCs) to skin tissues in vivo and discuss its effect on skin wound healing.Methods MSCs were isolated from porcine marrow,purified and cultured with 5-bromodeoxyuridine (5-BrdU) as a label in vitro.Forty-eight skin wounds on the six procines were divided into bFGF+MSCs(Group A),MSCs (Group B),bFGF (Group C) and control group randomly and treated correspondingly.Wound closing index (WCI),cell morphology and histological changes in the defected regions were evaluated 3,7,14,21 and 42 days after treatment.Vessel endothelium cells derived from MSCs in the granulation tissues were assayed by immuno-staining of 5-BrdU and Factor Ⅷ in the same cell.Results Healing speed of defected regions in Group A was faster than that of the other groups.Hierarchical scores of histopathology on the specimens of Group A were also higher than that of the other groups.The dual-positive stained cells by BrdU and Factor Ⅷ in Group A were more than those in Group B.Conclusion bFGF can induce MSCs differentiating into vessel endothelium cell and hence promote the repair of skin wounds.9 refs,3 figs,5 tabs.
