硝酸氧化纳米炭的制备及淋巴靶向的初步考察

摘要 目的制备出在分散介质中能够稳定存在的纳米炭混悬液,作为靶向至淋巴结的理想载体。方法采用硝基氧化的方法制备纳米炭,以扫描电镜、红外光谱仪和粒度测定仪表征制备的纳米炭,通过分散稳定性试验考察其在水性分散体系中的稳定性,并对纳米炭混悬液的淋巴靶向性进行了初步研究。结果制备出的纳米炭经表征初步判断在表面引入了硝基和羟基含氧官能团,在水性体系下的分散稳定性有显著的提高,平均粒径为80.5nm,并且具有较为理想的淋巴靶向和示踪效果。结论采用硝酸氧化制备的纳米炭能够稳定分散在水性体系中,具有较好的淋巴亲和性。 Objective To prepare nano-carbon suspension which has good stability in dispersion medium as an appropriate drug carrier to target regional lymph nodes.Methods Activated carbons were oxidized by nitric acid,which were characterized by means of SEM,FT-IR and particle-size analyzer,respectively.The dispersion stability of the nano-crabon was investigated using spectrometer.Pilot study of targeting nano-carbon suspension to lymph nodes was carried out.Results The results show that some marked hydrophilic functional groups on nano-carbon surfaces,including —NO2 and —OH,were increased greatly by nitric acid oxidation and the dispersion stability was improved significantly.The mean particle size of nano-carbon is 80.5 nm.The oxidized carbon nanoparticle suspension demonstrated good lymphatic targeting and tracing properties.Conclusion Oxidized nano-carbon suspension is stable and the lymphatic affinity is superior to original activated carbon.
