5-脂氧合酶在肿瘤中的作用和机制

摘要 5-脂氧合酶(5-lipoxygenase,5LO)是催化花生四烯酸(arachidonic acid,AA)转化为白三烯(LT)和氢氧化物5-羟基花生四烯酸(5-HETE)的关键酶。近年来,多聚不饱和脂肪酸代谢与肿瘤形成之间关系的分子学研究,为肿瘤的发生和发展提供了新的理论,也为肿瘤的预防和治疗提供了新的靶作用分子。大量研究发现抑制5LO能够抑制多种恶性肿瘤的增殖,并诱导细胞凋亡。现就5-脂氧合酶在肿瘤中的作用和机制作一综述如下。 5LO is a key enzyme to catalyze arachidonic acid which can transform into LT and 5-HETE.In recent years,the molecular studies about the relationship of poly-unsaturated fatty acid metabolism and tumor formation provide new theories to the tumor's occurrence and deveplopment,and also provide new molecular target to the tumor's precaution and therapy.Large number of studies found that 5LO can inhibit some kinds of malignant tumor's prolifetation,and induce apoptosis.will discuss The role and the mechanism of the 5LO in the tumor,are discussed in this paper.
