降脂膏对高脂大鼠血液流变学的影响

The Effects of Jiangzhigao on Hemorrheology of Rats With Hyperlipoidemia

摘要 目的:研究降脂膏(JZG)对高脂大鼠一氧化氮/内皮素、前列环素/血栓烷B_2及血液流变学的影响。方法:用高脂乳剂喂饲40d以形成大鼠高脂血症及动脉粥样硬化模型,同时给药组igJZG40、60、80/kg,测定血中NO、ET-1、6-keto-PGF_(1a)、TXB_2及血液流变学各指标。结果:JZG对正常及高脂大鼠血清NO水平有一定升高,明显降低ET-1水平,大剂量能显著升高、6-keto-PGF_(1a)含量,有利于维持6-keto-PGF_(1a)-PGF_1/ TXB_2平衡;可降低全血比粘度、全血还原比粘度、血浆比粘度、降低RBC聚集指数、压积,缩短RBC电泳时间,并能降低纤维蛋白原含量,对二磷酸腺苷诱导的正常大鼠血小板聚集,亦有一定的抑制作用。结论:JZG改善高脂大鼠血液流变学异常、恢复NO、ET-1、6-keto-PGF_(1a)、TXB_2平衡,可能是其抗动脉粥样硬化作用的机制之一。 Objective:To study the influences of Jiangzhigao(JZG)on NO/ET,6-keto-PGF_(1a)/TXB_2 and hemorrheology of rats with hyperlipidemia.Methods:The hyperlipidemia and early artherosclerosis model in rats were established by feeding high lipid diet for 40 days.Meanwhile JZG was taken by oral administration at the dosages of 40,60,80g/kg respectively;The levels of NO,ET-1,6-keto-PGF_(1a),TXB_2 in serum were determined,and the hemorrheology markers were observed.Results:JZG raised the levels of NO in serum,obviously decredsed ET-1 levels of both normal and the model rats.The large dose of JZG has remarkably increased 6-keto-PGF_(1a) content which benefits maintenance of the balance of 6-keto-PGF_(1a)/TXB_2,decreased the whole blood specific viscosity,whole blood reduction specific viscosity,plasma specific viscosity;aggregation index of RBC;shortened RBC electrophoresis time;also has decreased fibrinogen content;the platelet aggregation of normal rats induced by ADP was inhibited too.Conclusion:JZG can improve the abnormal hemorrheology and recover the balances of TXB/6-keto-PGF_(1a) and NO/ET of rats with hyperlipidemia,which might be one of mechanisms of antiartherosclerosis action.
