摘要
目的探讨大鼠脑缺血再灌注后海马CA1区神经元损伤的病理改变及三七总皂苷 (PNS)对其的影响。方法采用线栓法制备大鼠左侧大脑中动脉阻断 (MCAO)局灶性脑缺血模型 ,缺血 2h,再灌注 46h。大鼠随机分为假手术组、缺血再灌模型组、PNSⅠ组和PNSⅡ组。以神经症状缺损记分、海马CA1区组织学分级、海马CA1区神经元密度等作为评价指标。结果三七总皂苷组神经症状明显减轻或接近正常 ,其行为评分得分明显低 ;三七总皂苷组海马CA1区组织学分级降低、神经元密度增加 ,与模型组间有显著性差异。结论三七总皂苷能减轻脑缺血再灌注引起的损伤性神经症状及海马CA1区神经元损伤的程度。
Objective To investigate the pathological changes of neurons in hippocampus CA1 following cerebral ischemia-reperfusion in rats and the effects of Panax notoginseng saponins (PNS) on this kind of damage.Methods The model of local cerebral ischemia-reperfusion was prepared by the intravascular theread-occlusing procedures to cause 2 hours of occlusion and followed by 46 hours of reperfusion in rats.The animals were randomly divided into 4 groups, i.e.sham operation group (SOG), modeling group (MG),treating group 1 with PNS (TG1) and treating group 2 (TG2), also treated with PNS at different dosage.Then, the effects of PNS on the pathological changes were analyzed based on the evaluation of scoring on the neural defecting symptoms, histologial grading of the diseased area and the neuronal density in hippocampus CA1.Results Compared with that of MG, the neural defecting symptoms were significantly improved, or nearly similar to that of normals with wery lower behavior scores among the aminals of TG1 and TG2.Furthermore,reduced histological grades and increased neuronal density were also seen in TG1 and TG2,with significant differences between TGs and MG.Conclusion PNS holds definite protective effects on the damage caused by cerebral ischemia-reperfusion to improve the neural defecting symptoms and to reduce the degree of neural injuty.
