TLR4对人乳腺癌细胞MCF-7放射敏感性的影响

摘要 目的初步探讨Toll样受体(TLR4)对人乳腺癌MCF-7细胞放射敏感性的影响。方法将体外培养的MCF-7细胞分为假照组和5 Gy照射组,将其各自分为空白对照组、TAK242阻滞组和LPS刺激组,采用CCK-8试剂盒检测其增殖活性,用Annexin V凋亡试剂盒检测其凋亡率,用流式细胞术观察细胞周期进程的变化,用克隆形成实验计算其细胞存活分数。结果 5 Gy照射后MCF-7细胞的增殖活性明显降低(P<0.05),与对照组相比,TAK242阻滞TLR4后MCF-7细胞增殖活性更加降低(P<0.05),而LPS刺激TLR4后MCF-7细胞增殖活性明显升高(P<0.05)。MCF-7细胞受照后G2/M期细胞显著多于对照组(P<0.01),G0/G1期细胞明显减少(P<0.01),S期细胞变化不明显(P>0.05)。5 Gy照射后MCF-7细胞的凋亡率明显高于假照组(P<0.01),与对照组相比,阻断TLR4后其凋亡率明显升高(P<0.05),而激动TLR4后其凋亡率明显降低(P<0.05)。5 Gy照射后MCF-7细胞的存活分数明显降低(P<0.05),与对照组相比,阻滞TLR4后MCF-7细胞存活分数明显降低(P<0.05),而激动TLR4后MCF-7细胞存活分数显著升高(P<0.05)。结论 TLR4能够增强人乳腺癌MCF-7细胞的辐射敏感性,可致细胞G2期阻滞,但其机制需要进一步探讨。
