摘要 目的探索去卵巢对大鼠Wnt/β-catenin信号通路的影响。方法将20只3月龄SD雌性大鼠随机分为去势组和正常对照组,每组10只。去势组大鼠行双侧卵巢切除术,正常对照组大鼠不作任何处理。去除卵巢12 w后,取血检测血清雌二醇、骨源性碱性磷酸酶和抗酒石酸酸性磷酸酶水平;取左股骨进行骨密度检测;取右股骨骨髓检测低密度脂蛋白受体相关蛋白5(LRP5)和β-catenin mRNA和蛋白表达。结果去势组大鼠血清雌二醇水平较正常对照组显著下降(P<0.01),骨源性碱性磷酸酶和抗酒石酸酸性磷酸酶水平较正常对照组显著增高(P<0.01);去势组大鼠骨密度比正常对照组显著降低(P<0.01);去势组的β-cantenin mRNA和蛋白表达比正常对照组显著增高(P<0.05),尽管去势组的LRP5 mRNA和蛋白表达比正常对照组增高,但差异没有统计学意义。结论大鼠去卵巢12 w后Wnt/β-catenin信号通路被激活。 Objective To explore the effects of ovariectomy on Wnt/β-catenin signaling pathway in rats.Methods Twenty three-month-old female SD rats were randomly divided into two groups:normal control and the ovariectomized groups.Bilateral ovariectomy was performed on rats in the ovariectomized group.After 12 weeks, serum 17β-oestradiol, bone specific alkaline phosphatase and tartrate-resist-ant acid phosphatase 5b were analyzed by using ELISA.Bone mineral density of the left femur was measured using DEXA scan.mRNA ex-pressions of low density lipoprotein receptor related protein 5 (LRP5)andβ-catenin were evaluated using RT-PCR, and protein expressions were evaluated using Western blot.Results Serum 17β-oestradiol level was significantly lower , while bone specific alkaline phosphatase and tartrate-resistant acid phosphatase 5b levels were significantly higher in the ovariectomized group than that in normal control group ( all P<0.01 ).Bone mineral density was significantly lower in the ovariectomized group than that in normal control group ( P<0.01 ).mRNA and protein expression of β-catenin in the ovariectomized group were significantly higher compared with that of normal control group ( P<0.05 ).Though mRNA and protein expressions of LRP 5 increased in the ovariectomized group , the difference was not statistically significant from that in normal control group.Conclusions Ovariectomy has a significant impact on Wnt/β-catenin signaling pathway in rats.
