摘要
目的:探讨天麻素对S100β表达的影响,明确天麻素的神经保护机制。方法采用线栓法阻断大脑中动脉(MCAO)2 h诱导建立暂时性局部脑缺血模型,术前连续3 d给予 SD 大鼠腹腔注射天麻素,免疫组织化染色观察缺血边缘区 S100β阳性细胞数目变化;ELISA 检测血清S100β的浓度。结果天麻素预处理组大鼠S100β阳性细胞数目增加较少,1、3 d与缺血组和生理盐水组比较均有显著差异( P<0.05)。 ELISA测定结果显示MCAO后血清S100β含量1 d时迅速升高,3~7 d成倍数下降。天麻素处理组1、3 d大鼠血清S100β浓度明显降低(P<0.05)。结论天麻素能明显抑制脑缺血再灌注后S100β的过度表达,减轻神经细胞损伤。
Objective To explore the effect of Gastrodin on expression of S100β after cerebral post-ischemic reperfusion. Methods Gastrodin was intraperitoneally injected daily for 3days before inducing focal cerebral ischemia in SD rats by intraluminal occlusion of middle cerebral artery( MCAO) for 2 hours. Immunohistchemical method was used to observe the expression of S100β. The concentration of S100β in serum was detected by double-antibodies-sandwich-ELISA method as well. Results After MCAO,the number of S100β positive cells in gastrodin group was significantly decreased compared with the ischemia group at 1 day and 3 day respectively( P<0. 05). The concentrations of S100β in serum remarkablely decreased campared with the ischemia group at 1 and 3 days( P < 0. 05) in Gastrodin group. Conclusions Gastrodin could inhibit the over-expression of S100β,which may be one of the neuroprotective mechanism of Gastrodin.
