摘要 目的:研究大豆异黄酮对体外培养的大鼠睾丸支持细胞(sertoli cells,SCs)增殖以及卵泡雌激素受体(folliclestimulating hormone receptor,FSHR)、抑制素α亚基(inhibinα,INHα)、抑制素βB亚基(inhibinβB,INHβB)、雄激素结合蛋白(androgen binding protein,ABP)、转铁蛋白(transferrin,Tf)和波形蛋白(vimentin)mRNA水平的影响。方法:体外培养大鼠SCs,用0.03,0.3,3及30μmol/L的大豆黄酮(daidzein,Dai)和0.05,0.5,5及50μmol/L的染料木黄酮(genistein,Gen)处理细胞,MTT检测细胞增殖,实时荧光定量PCR检测FSHR,INHα,INHβB,ABP,Tf和vimentin mRNA的相对表达量。结果:与对照组相比,其余各组细胞增殖和INHβB,ABP mRNA相对表达量差异均无统计学意义(P>0.05),Dai(0.3及3μmol/L)和Gen(0.05μmol/L)组INHαmRNA表达增加,Dai(30μmol/L)和Gen所有浓度组的FSHR mRNA表达下降,Dai(30μmol/L)和Gen(5μmol/L和50μmol/L)组Tf mRNA表达下降,Dai(3μmol/L及30μmol/L)和Gen(50μmol/L)组vimentin mRNA表达下降。结论:大豆异黄酮可能通过抑制SCs重要功能蛋白的mRNA转录而影响其功能发挥,从而对雄性生殖系统产生负面影响。 Objective: To determine the effect of soy isoflavones on cell proliferation and the transcription levels of follicle-stimulating hormone receptor(FSHR), inhibin α(INHα), INHβB, androgen binding protein(ABP), transferrin(Tf) and vimentin in testis sertoli cells in SD rats. Methods: Sertoli cells were cultured in vitro, exposed to daidzein at 0.03, 0.3, 3, and 30 μmol/L and genistein at 0.05, 0.5, 5 and 50 μmol/L, respectively. MTT was used to detect the proliferation of sertoli cells. Real-time PCR was used to detect the relative mRNA expressions of FSHR, INHα, INHβB, ABP, Tf and vimentin.Results: Compared with control groups, cell proliferation and the relative mRNA expression levels of INHβB and ABP in the treated cells showed no significant alternation. The INHα mRNA expression levels were increased in 0.3 and 3 μmol/L Dai and 0.05 μmol/L Gen, while the mRNA expression levels of FSHR were downregulated in 30 μmol/L Dai and Gen at all concentrations. Tf mRNA expression levels were downregulated in 30 μmol/L Dai and 5 μmol/L and 50 μmol/L Gen, and the mRNA expression levels of vimentin were downregulated in 3 and 30 μmol/L Dai and 50 μmol/L Gen. Conclusion: Soy Isoflavones may have potential detrimental effect on the male reproductive system, as they may impact the function of sertoli cells by downregulating the transcription levels of some important proteins.
