摘要 目的建立光纤生物传感器快速检测沙门菌的方法,为沙门菌的检测提供一种快速、简便、准确的实时检测方法。方法采用双抗体夹心免疫反应模式,优化捕获抗体和检测抗体浓度,在最优实验条件下确定方法的灵敏度和特异度,同时检测环境水样验证方法的灵敏度和特异度。结果该方法检测沙门菌的检出限为5.5×103cfu/mL,完成一次完整的检测用时为40 min,对沙门菌代表性血清型的低浓度菌液进行检测均为阳性,对非沙门菌的检测结果均为阴性。结论光纤生物传感器检测沙门菌具有快速、灵敏、特异度好等优点,可作为快速检测沙门菌的一种方法。 Objectives To establish a fiber optic biosensor method for rapid,convenient and accurate detection of Salmonella. Methods Applying a double-antibody sandwich immunoassay mode and optimizing the concentration of capturing and detecting antibody,the sensitivity and specificity were determined under the optimum conditions,and the sensitivity and specificity of the method was validated through detecting the environmental water samples. Results The detection limit of fiber optic biosensor method for Salmonella was 5. 5 × 103cfu/mL,and the response time was about 40 min. The detection results for Escherichia Coli,Staphylococcus aureus,Shigellaflexneri and Shigelladysenteriae were all negative and showed a favourable specificity. Conclusions The advantages of fiber optic biosensor method were rapid,sensitive and specific in the detection of Salmonella. It can be used as a convenient method for Salmonella detection.
