摘要
目的确定瓜蒌桂枝解痉颗粒的最佳提取工艺。方法采用正交试验设计、响应面法,以桂皮醛含量、干膏得率为考察指标,考察浸泡时间、提取时间、提取次数、料液比、减浓缩温度对提取效果的影响,在单因素分析的基础上选择对响应值有显著影响的因素,采用正交试验设计、响应面法比较提取工艺条件。结果瓜蒌桂枝解痉颗粒响应面优化后的提取工艺条件为:料液比1∶10,药材浸泡38 min,煎煮2次,每次20 min,减压浓缩温度62.4℃;桂皮醛含量与干膏得率预测值与实测值的偏差分别为2.11%和1.79%。结论响应面法所得结果更精确,正交设计试验次数少,方法简便。
Objective To study the optimum extraction process of Gualou Guizhi Jiejing granules.Methods The content of cinnamic aldehyde and dry extract yield considered as evaluation indexes were determined by orthogonal design and response surface optimization, and the effects of soaking time, extracting time, extracting times, ratio of solid to liquid and decompression and concentration temperature on extracting result were investigated. The factors which had significant effects on response value were choosed based on single factor analysis, and the two kinds of technologcial conditions were compared. Results After using response surface optimization, the optimal conditions were as follows:ratio of solid to liquid was 1:10, soaking time was 38 minutes, decocting times was 2 and each time for 20 minutes, decompression and concentration temperature was 62.4℃, and deviation of predictive value and practical value of cinnamic aldehyde content,dry extract yield was 2.11%, 1.79% respectively. Conclusion The result of response surface method is more accurate. The orthogonal design which needs fewer test is simple.
