摘要
目的研究小鼠支气管哮喘模型不同时间段IFN-γ、IL-4、IL-17浓度的变化,探讨其在哮喘发病机制中的作用。方法卵白蛋白(OVA)致敏建立小鼠哮喘模型,BALB/c小鼠40只,随机分为3组:正常对照组8只,模型对照组8只,哮喘模型组24只。哮喘模型组分别在雾化攻击6d、10d及14d(分别称为哮喘6d、10d、14d组)后随机选取8只小鼠处理,取各组小鼠支气管肺泡灌洗液(BALF)及血清,测定IFN-γ、IL-4、IL-17的浓度。结果①.与健康对照组相比,哮喘模型组(包括6d、10d、14d)BALF及血清中的IL-4和IL-17水平升高,而IFN-γ水平则降低,差异具有统计学意义(P<0.05);②.BALF中14d哮喘组各细胞因子水平与6d哮喘组比较,差异也具有统计学意义(P<0.05),但10d哮喘组和6d哮喘组间无明显差异(P>0.05);③.不同时间段哮喘模型血清中细胞因子水平没有差异(P>0.05)。结论 IFN-γ、IL-4、IL-17在哮喘模型不同发病阶段水平会发生变化,因此这些细胞因子在哮喘的发生发展中可能发挥了一定的作用。
Objective To investigate the changes of IFN-γ,IL-4and IL-17in asthmatic mice of different time stages and analyze the significance.Methods The mouse models of asthma were established by sensitization with ovalbumin(OVA).40BALB/c mice were divided into three groups including asthma model(24),normal control(8)and model control groups(8).8asthmatic mice were chosen separately after 6d,10dand 14dof inhalation,and concentrations of IFN-γ,IL-4and IL-17in serum and bronchoalveolar lavage fluids(BALF)were detected by ELISA.Results 1.Compared with those of normal control group,concentrations of IL-4and IL-17in serum and bronchoalveolar lavage fluids(BALF)from asthma model group(include 6d,10dand 14d)were increased,while IFN-γshowed a decrease,and the differences were statistically significant(P<0.05).2.In the BALF,cytokines from 14dasthma model group and 6dmodel group also showed statistical difference(P<0.05),but there was no distinct difference between 6dand 10dmodel group(P>0.05).3.The concentration of cytokines in serum from different time stages of asthmatic model showed no distinct difference(P>0.05).Conclusion The concentrations of IFN-γ,IL-4and IL-17were changed in different time stages of mouse asthma model,and also,they may play a role in the development of asthma.
