The Transcription of β-arrestin2 mRNA in Different Encephalic Regions of Rats Anesthetized with the Combined Anaesthetic for Miniature Pigs

摘要 为研究小型猪专用复合麻醉剂(XFM)对大鼠不同脑区β-arrestin2mRNA转录的影响,将30只大鼠随机分为对照组(C组)和麻醉组(M组),M组分为4个亚组:M1组(注射XFM后大鼠翻正反射消失即刻)、M2组(注射XFM后大鼠翻正反射消失后1h)、M3组(注射XFM后大鼠翻正反射恢复即刻)和M4组(注射XFM后大鼠翻正反射恢复后1h),各组大鼠到达预定的时间点后分别采取脑组织,应用实时荧光定量PCR技术检测各组织中β-arrestin2mRNA转录量。结果显示,大鼠经过XFM麻醉后,大脑皮层、小脑β-arrestin2mRNA的相对转录量在麻醉期间受到明显的抑制,与对照组相比差异极显著(P<0.01),海马和脑干β-arrestin2mRNA的相对转录量在麻醉期间明显增加,与对照组相比差异极显著(P<0.01)。结果表明,XFM能影响中枢神经系统中β-arrestin2mRNA的转录,为继续探索麻醉对MAPK通路的影响奠定基础。 In order to investigate the Transcription ofβ-arrestin2mRNA in different encephalic regions of rats anesthetized with the Combined Anaesthetic for Miniature Pigs(named as XFM),30 rats were divided randomly into two groups:the control group and XFM groups.XFM groups consisted of four subgroups:M1subgroup(rats received XFM intraperitoneally and then waiting the disappearance of right reflection);M2subgroup(rats received XFM intraperitoneally and then waiting for 1hafter the disappearance of right reflection);M3subgroup(rats received XFM intraperitoneally and then waiting the recovery of right reflection)and M4subgroup(rats received XFM intraperitoneally and then waiting for 1hafter the recovery of right reflection).The brain tissues were taken when reaching each time point.The transcription ofβ-arrestin2mRNA in central nervous system was detected with real time PCR.The results showed that the transcription of β-arrestin2mRNA in cerebral cortex and cerebellum decreased significantly after receiving XFM(P<0.01);the transcription ofβ-arrestin2mRNA in hippocampus and brain stem increased significantly(P<0.01).These results indicated that XFM could affect the transcription ofβ-arrestin2mRNA in central nervous system.This might be one of the anaesthetic mechanisms forXFM.And it will lay the foundation of continuing to explore anesthesia for the effects of MAPK pathway.
