摘要
目的初步探讨热疗对食管癌细胞株EC-1放射增敏作用及其可能机制,为热疗与放疗联合治疗食管癌提供理论依据。方法采用平板克隆形成分析法计算各组细胞存活分数SF,"多靶单击数学模型"拟合细胞存活曲线。流式细胞术检测细胞凋亡及细胞周期的变化。结果热疗对食管癌细胞株EC-1放疗有显著的增敏作用,热疗30 min的放射增敏比为1.060。热疗可使细胞阻滞在G2/M期,使G0/G1期、S期细胞比例减少,增加细胞凋亡,与对照组相比差异有统计学意义(P<0.05)。结论热疗对食管癌EC-l细胞具有增敏作用,热疗可通过影响食管癌EC-l细胞周期,诱导细胞凋亡,增强放射线对细胞的杀伤作用。
Objective To investigate the radiosensitizing effect and underlying mechanism of hyperthermia on human esophageal carcinoma cell line EC-1. Methods The radiosensitizing effect of hyperthermia human on esophageal carcinoma cell line EC-1 by fi tting the dose-survival curving‘multitarget click mathematical model'.Flow cytometry(FCM) was used to detect the changes of cell apoptosis and cell cycle.Results Hyperthermia had an obvious radiosensitization on human esophageal carcinoma EC-1 cells. After treatment with hyperthermia for 30 minutes, the sensitization enhancement ratio was 1.060,the G2/M phase ratio increased while S phase and G0/G1 phase decreased. The apoptosis ratio increased significantly(P<0.05). Conclusion Hyperthermia can enhance the radiosensitivity, and combination of them could enhance it more signifi cantly. Hyperthermia can enhance the killing effect of radiation on tumour cells mainly through affecting the EC-l cell cycle, and the induction of apoptosis is the second reason.
