DAPT通过抑制NOTCH信号通路对食管癌细胞株体外增殖的影响

摘要 目的探讨gama分泌抑制药物γ-分泌酶抑制剂(DAPT)通过抑制NOTCH信号通路对食管癌细胞株HE-1体外增殖的影响。方法将食管癌细胞随机分为四组,对照组和低、中、高实验组,实验组加入终浓度为1、5、10μmol/L的gama分泌抑制药物DAPT,对照组加入相应DAPT溶剂,检测细胞选择加入DAPT后的第0、24、48、72 h的细胞。癌细胞的增殖活性采用噻唑蓝(MTT)法进行检测,Notch信号通路活动情况采用Western印迹法检测相关蛋白的表达变化。结果三个剂量组72 h的抑制率分别为21.8%、53.0%、67.2%,高、中剂量组抑制率明显高于低于低剂量组(P<0.05);细胞增殖活性随DAPT接触时间的延长呈逐渐减低增强的趋势(P<0.05)。NOTCH信号通路中Notch2表达相对灰度值分别为0.801、0.615、0.410;hes1分别为0.812、0.623、0.453,组间差异有统计学意义(P<0.05)。蛋白的表达灰度值随DAPT接触时间延长呈逐渐减低增强的趋势(P<0.05)。结论 gama分泌抑制药物DAPT可以抑制NOTCH信号通路,进而对食管癌细胞株HE-1体外增殖发挥抑制作用。
