冠盖藤对骨性关节炎大鼠细胞因子的影响

Effect of Pileostegia viburnoides Hook.f.et Thorns on osteoarthritis cytokines

目的研究冠盖藤对骨性关节炎(OA)大鼠血常规及血清中白细胞介素1β(IL-1β)、肿瘤坏死因子α(TNF-α)、IL-6水平的影响。方法采用弗氏完全佐剂制备OA模型,将随机造模成功后大鼠随机分为模型组,雷公藤组,冠盖藤高、中、低剂量组。经药物干预28 d后,检测血常规及血清细胞因子水平。结果与正常组比较,模型组的血常规中白细胞、中性粒细胞数明显升高,血清IL-1β、IL-6、TNF-α的水平出现升高,经冠盖藤干预后血常规中白细胞、中性粒细胞数明显下降,血清中IL-1β、IL-6、TNF-α的水平明显降低,并且以高剂量组最明显。结论冠盖藤的抗炎镇痛机制之一可能通过降低白细胞、中性粒细胞数,血清IL-1β、IL-6、TNF-α水平。

Objective To study effect of Pileostegia viburnoides Hook.f.et Thorns on blood count and levels of serum interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α),interleukin-6(IL-6) in osteoarthritis rat.Methods Osteoarthritis model was made by Freund's Complete Adjuvant(FCA),then osteoarthritis model rats were randomly divided into model,tripterygium,high-,middle-,low-dose of Pileostegia viburnoides Hook.f.et Thorns.Rats were detected blood and serum cytokine levels after intervention 28 days later.Results Compared with the normal group,blood leukocytes,neutrophils,serum levels of IL-1β,IL-6,TNF-α were elevated in the model group.The number of blood leukocytes,neutrophils were decreased,the serum level of IL-1β,IL-6,TNF-α were significantly reduced in Pileostegia viburnoides Hook.f.et Thorns group,and the effects of high dose of Pileostegia viburnoides Hook.f.et Thorns were the most obvious.Conclusions One of the anti-inflammatory and analgesic mechanism may reduce leukocyte,neutrophil count,serum levels of IL-1β,IL-6,TNF-α.
