摘要
目的观察urantide对动脉粥样硬化(AS)大鼠肝脏的影响,探讨其防治AS的作用机制。方法采用高脂饲料喂养及腹腔注射维生素D3损伤动脉内膜的方法建立大鼠动脉粥样硬化模型,随机分为正常组、AS组、阳性药组、urantide组,通过HE染色方法观察各组肝脏的病变情况。结果 AS组肝系数明显增加(P<0.01);urantide组肝系数较AS组明显减少(P<0.01)。AS组大鼠肝脏脂肪变性细胞数≥50%,为Ⅳ级重度脂肪变性;而urantide组肝细胞脂肪变性逐渐减少,变形细胞数≥25%(<50%),为Ⅲ级中度脂肪变性,接近阳性药组水平。结论 urantide对AS大鼠肝脏有一定的保护作用,为临床应用urantide治疗AS提供实验依据。
Objective Methods To observe the influence of urantide on the expression of liver in rats with atherosclerosis( AS) and investigate its mechanism. Results AS model was induced by fed high fat diet and arterial intimal injury of intrape ritoneal injection of vitamin D3,rats were randomly divided into four groups,hematoxylin-eosin staining was used to observe the change of the liver situation. Results: The hepatic index of AS group was increased remarkably( P < 0. 01). Compared with AS group,the hepatic index of urantide group was reduced significantly( P < 0. 01). The number of rats liver steatosis cells of AS group was ≥50%,Ⅳ level severe fatty degeneration; but hepatic steatosis of urantide group was reduced gradually,the number of deformation cells was ≥25%( < 50 %),and moderating fatty degeneration,closing to the level of positive medicine group. Conclusions Urantide has significant protective effect against liver in AS rats,which provides new views and experimental basis for the clinic treatment of AS with urantide.
