摘要
目的探讨复方地黄方对帕金森病(PD)左旋多巴毒副反应大鼠淋巴细胞(PBL)表面多巴胺转运体(DAT)表达的影响。方法在经典注射6-羟基多巴胺损毁法制作PD大鼠模型的基础上,进一步腹腔注射50 mg·kg-1·d-1左旋多巴(LD)/苄丝肼2 w,制备LD大鼠模型。采用随机区层分组法,分为正常对照组、假手术组、LD模型组及复方地黄方组四组,每组12只,分别干预4 w、6 w,应用RT-PCR法测定大鼠PBL表面DAT的表达。结果 LD模型组大鼠PBL表面DAT的表达较正常及假手术组明显减少(P<0.001),且与病程成负相关;复方地黄方组在不同时间点均明显高于LD模型组(P<0.001)。结论复方地黄方可能通过增加PBL表面DAT表达来发挥减毒抗PD的作用。
Objective To study the effects of compound rehmannia formula on expression of Dopamine transporter( DAT) DAT mRNA content in the peripheral blood lymphocyte( PBL) of Parkinson disease( PD) rats treated with L-dihydroxy-phenylalanine( LD) levodopa. Methods On the basis of PD rat model with targeted injection of 6-OHDA in unilateral brain,and further intraperitoneal injection of 50 mg·kg-1·d-1LD / benserazide for 2 weeks to establish LD rat PD model. The rats were randomly divided into the normal control,sham,LD treatment and the compound rehmannia formula groups( n = 12). According to different intervention time points,each group was divided 4, 6 weeks groups( n = 6). RT-PCR was used to detect DAT mRNA expression on the peripheral blood lymphocyte surface. Results Compared with the normal control and sham groups,expression of DAT on PBL of the damaged part in LD treatment group were significantly decreased( P < 0. 001),and had a negative correlation with the course. Compared with LD treatment group,expression of DAT on PBL of the compound rehmannia formula group were significantly increased during different phases( P < 0. 001). Conclusions Compound rehmannia formula can reduce the adverse effects of LD to treat PD by increasing the expression of DAT mRNA on PBL.
