按摩对兔骨骼肌钝挫伤炎症及血液机制的影响 被引量:7

摘要 目的研究新西兰兔股四头肌经重物外力损伤后,在肌组织损伤修复过程中,按摩对炎症因子IL-6、PGE2、CRP及血液流变学的影响以及超生影像学和组织学的改变,探讨按摩促进肌肉损伤修复的作用机制。方法健康成年雄性新西兰大白兔40只,体重(2.0±0.5)kg,随机分为4组,正常对照组(A组,n=4)、按摩前对照组(B组,n=4)、自然恢复组(C组,n=16)和按摩组(D组,n=16)。A组实验动物不作任何处理,作为正常对照;B、C、D组实验动物用自制打击器制备兔右后肢股四头肌损伤模型。D组于造模后第5天开始按摩〔5〕,C组不予按摩。C、D组于伤后7、11、15及19 d各取4只实验动物取股四头肌样本,用酶联免疫吸附法检测以上各组血清中IL-6、PGE2、CRP的含量,同时测量血液流变学的变化;HE染色及微泡超声影像技术检测观察制模后7、15 d的组织病理变化。结果按摩干预后,按摩组与自然恢复组比较,PGE2、IL-6、CRP含量下降显著(P<0.01),全血黏度与血浆黏度均显著降低(P<0.01)。超声微泡造影显示,D组较C组损伤部位较轻,血供较丰富,坏死区明显减轻,且血管恢复较快。结论按摩能通过调节IL-6、PGE2、CRP炎症因子含量,减轻炎症反应,改善血流状态,促进血管生成,从而修复受损肌组织。 Objective To investigate the effect of massage on quadriceps femoris repair after injury by external force and the influ- ence of inflammatory markers IL-6,PGE2,CRP and blood rheology and the ultrasonic imaging and histological observation in New Zealand rabbits,so as to explore the possible molecular mechanisms of massage in repair of muscle injury.Methods Forty New Zealand white rab- bits,weighting(2.0 ± 0.5) kg,were randomly divided into normal control(A)(n = 4),the control group before the massage(B)(n = 4),self-repairing(C)(n = 16,further divided into 7,11,15,19 d),and massage groups(D)(n = 16,further divided into 7,11,15,19 d).In group A,the rabbits were not treated as normal controls; in groups B,C and D,the rabbit models of quadriceps femoris injury were prepared by self-made beater.In group C,no massage therapy was given as nature recovery controls; in group D,massage therapy was given after 5 d since model built successful as massage group.C and D groups after injury in 7,11,15,19 d each take 4 different animals were taken quad- riceps samples.The contents of IL-6,PGE2,CRP and the changes of hemorrheologic were detected by ELISA,the organization pathological changes were observed by Ultrasound images technical and HE staining.Results After massage intervention,the contents of PGE2,IL-6,CRP in massage group were decreased significantly compared with those of control group(P<0.01),and blood rheology displayed that the whole blood viscosity and plasma viscosity were significantly decreased(P<0.01).Ultrasound of micro-bubble angiography showed that the injury parts was lighter in unmassaged(C) group than that of massaged group(D),blood supply was richer,necrotic area was significantly decreased,and vascular restore was faster.Conclusions Massage could adjust the content of inflammation factors such as IL-6,PGE2 and CRP,reduce inflammation reaction,improve blood flow state,promote angiogenesis,repair damaged muscle tissue.
