17-β-雌二醇对去卵巢胰岛素抵抗大鼠骨骼肌中葡萄糖转运体4表达的影响 被引量:2

摘要 目的观察17-β-雌二醇对高果糖诱导的去卵巢胰岛素抵抗大鼠骨骼肌中葡萄糖转运体4(GLUT4)表达的影响。方法 40只成年雌性SD大鼠随机地分为正常对照组、模型组、17-β-雌二醇替代组和溶媒对照组。模型组大鼠卵巢切除后,高果糖饲料喂养8 w诱导胰岛素抵抗的产生;17-β-雌二醇替代组大鼠卵巢切除后,高果糖饲料喂养8 w同时给予17-β-雌二醇(30μg/kg)每天皮下注射。溶媒对照组大鼠给予相同剂量的乙醇。结果与正常对照组相比,模型组大鼠体重、收缩压、空腹血糖、血清胰岛素、内脏脂肪重量、甘油三酯、总胆固醇和低密度脂蛋白均显著增加,高密度脂蛋白、股四头肌糖摄取和胰岛素敏感指数均显著降低,股四头肌中p-Akt和GLUT4 mRNA和蛋白的表达显著性降低;17-β-雌二醇替代逆转上述改变。结论 17-β-雌二醇抑制高果糖诱导的去卵巢大鼠胰岛素抵抗,其机制与17-β-雌二醇上调骨骼肌中p-Akt和GLUT4的表达有关。
