红细胞膜软骨寡聚基质蛋白抗原抗体力学检测在膝骨关节炎早期诊断中的作用 被引量:2

摘要 目的运用原子力显微镜(AFM)探针修饰上软骨寡聚基质蛋白(COMP)抗体检测骨关节炎(OA)高危人群、OA早期患病组红细胞膜上COMP抗原间相互作用力,探讨这种方法在OA早期诊断中的作用。方法对62例单侧膝关节OA早期患者、9例OA高危人群及8名正常人抽取静脉血,采用ELISA法测定血清样本中COMP的水平。同时分离红细胞,运用AFM探针修饰上COMP抗体检测与红细胞膜上抗原的相互作用力。结果 OA早期患者血清中COMP含量明显高于正常人和OA高危人群(P<0.05),但OA高危人群与正常人差异无统计学意义(P>0.05)。COMP血清学检测无法区分OA高危人群与正常人。OA早期患者、正常人、OA高危人群红细胞膜COMP黏附力和弹性模量差异均有显著差异。OA高危人群红细胞COMP力学性能已经明显增加,当发生OA时,黏附力和弹性模量参数达到峰值。结论运用AFM探针修饰上COMP抗体检测与红细胞膜上抗原的相互作用力可以作为临床早期诊断OA的特异性方法,可以作为OA高危人群的初筛。
