摘要
目的探讨寡核苷酸微阵列芯片技术在人乳头瘤病毒(HPV)检测中的实用价值。方法使用HPV寡核苷酸微阵列芯片试剂盒对227例宫颈拭子样本进行16种高危型(HPV-16,18,31,33,35,39,45,51,52,53,56,58,59,66,68,CP8304)和5种低危型(6,11,42,43和44)检测,所检测的样本为HC2探针捕捉法检测所得到的HPV高危型和低危型样本,用以评估该DNA微阵列试剂的性能。结果190例经HC2法检测为高危型HPV阳性以及30例被HC2法检测为低危型的样品被寡核苷酸微阵列芯片试剂盒检测出具体的高危和低危HPV型别,总共检测出13种高危型和5种低位危型HPV亚型,在高危亚型中,各型检出率分别是16型26.01%、58型20.18%、52型14.71%、68型7.59%、31型7.13%、33型6.37%、66型6.24%、18型4.05%、59型2.18%、39型1.10%、56型1.50%、45型1.40%、51型1.07%;在低危亚型中,各型检出率分别为6型45.43%、11型29.70%、42型14.11%、43型5.89%和44型5.00%。单一HPV亚型感染及2种、3种HPV亚型混合感染分别为60.46%、29.89%、9.65%。7例经HC2检测为高危型,但寡核苷酸微阵列芯片试剂盒未能检测出HPV的具体型别,寡核苷酸微阵列芯片试剂盒与HC2法高危型HPV检测结果的符合率为96.92%(220/227)。结论HPV寡核苷酸微阵列芯片试剂盒与HC2法高危型HPV检测阳性符合率高,并可以明确具体分析21种HPV型别,是一种准确、高效、经济的HPV型别检测方法,可用于HPV的常规基因型分型诊断及人群筛查。
Objective To explore the application value of oligonucleotide micoarray for human papillomavirus(HPV) detection.Methods 227 samples of dacron swabs are tested by Oligonucleotide Micoarray kit for HPV genotyping of 16 high risk subtypes(HPV-16,18,31,33,35,39,45,51,52,53,56,58,59,66,68,CP8304),and 5 low risk subtypes(HPV6,11,42,43,44),the samples were HC2 kit tested for high risk and low risk subtypes,be intended for performance evaluation of the Oligonucleotide Micoarray kit.Results 190 high risk and 30 low risk HPV subtype positive cases were genotyped sepciifcally by Oligonucleotide Micoarray kit via blind assay.13 high risk HPV genotypes were detected,namely HPV 16(26.01%),58(20.18%),52(14.71%),68(7.59%),31(7.13%),33(6.37%),66(6.24%),18(4.05%),59(2.18%),39(1.10%),56(1.50%),45(1.40%),51(1.07%),and 5 low risk HPV genotypes were detected as 6(45.43%),11(29.70%),42(14.11%),43(5.89%) and 44(5.00%).The detection rates of 1 single HPV genotype infection and 2,3 HPV genotypes mixed infection were 60.46%,29.89%,9.65% resepctively.7 samples were not detected for genotype.The coincidence rate was 96.92% between Oligonucleotide Micoarray kit and HC2.Conclusion Oligonucleotide Micoarray kit was a rapid and economic method to identify HPV genotype and high coincidence rate with HC2 and could be applied for HPV genotyping and screening of HPV infection.
