藻蓝蛋白对2型糖尿病大鼠心肌细胞凋亡影响的研究

The effect of phycocyanin on cardiocytes apoptosis in rat models with type 2 diabetic mellitus

摘要 目的:研究藻蓝蛋白对2型糖尿病大鼠心脏形态结构和心肌细胞凋亡的影响。方法:健康雄性Wistar大鼠32只,高脂高糖饲料喂养联合小剂量链脲佐菌素(STZ)腹腔注射方法建立2型糖尿病大鼠模型,藻蓝蛋白灌胃治疗,组织病理学技术观察心脏的形态结构,用原位末端标记法(Tunel法)检测心肌细胞凋亡。结果:正常组大鼠心肌纤维走行和结构正常,心脏中均可见少量散在的棕黄色染色的凋亡阳性心肌细胞;模型组大鼠心肌纤维肥大、颗粒变性、脂肪变性,凋亡阳性细胞较正常组明显增多,凋亡指数有显著差异(P<0.05);藻蓝蛋白组和二甲双胍组心肌细胞形态结构较模型组显著改善,正常细胞数目增多、排列较规则,肿胀和变性细胞较模型组显著减少,凋亡阳性细胞较模型组显著减少,凋亡指数有显著差异(P<0.05)。藻蓝蛋白组和二甲双胍组凋亡指数无显著性差异(P>0.05)。结论:藻蓝蛋白可抑制糖尿病大鼠心肌细胞凋亡,发挥一定的保护作用。 Objective:To investigate the effect of phycocyanin on the morphosis and apoptosis of cardiocytes in rats with type 2 diabetic diseases. Methods:Type 2 diabetes mellitus models were induced by high fat and sugar diet and low dose intraperitoneal injection of streptozotocin (STZ) in 32 healthy male Wistar rats, and treated with phycocyanin by intragastric administration. The morphosis of heart was observed by histopathological technique and the apoptosis of cardiocytes were detected by Tunel assay. Results : There were a few yellow positive apoptotic cells in heart and the cardiac muscle fibers courser and structured normally in normal group. In the model group, the cardiac muscle fibers swelling, granular and fatty degeneration, and the number of positive apoptotic cells and apoptosis index were significantly higher than those in normal group (P<0.05). In phycocyanin and dimethylbiguanide group, the morphosis of heart improved significantly and the number of normal cells increased regularly with decreasing of swelling and degenerative cells, and the the number of positive apoptotic cells and apoptosis index were significantly lower than those in model group (P<0.05). There was no significant difference between phycocyanin and dimethylbiguanide groups (P>0.05). Conclusion:Phycocyanin might play a protective effect by inhibiting cardiocytes apoptosis in diabetes mellitus rats.
