同种异体肌腱不同时段深低温保存对肌腱愈合影响的实验研究 被引量:2

Experimental Study of the Allogenic Tendon Preservation in Different Time Interval in Deep Low Temperature Influencing on Tendon Healing

摘要 目的:探讨深低温(-80℃)保存异体肌腱的最佳时间段。方法:体重250-300g的雌雄不分的SD大鼠144只,随机分为Ⅰ、Ⅱ两组。每组72只。Ⅰ组为供腱组,提供144条供移植用的肌腱。Ⅱ组又随机分为A、B、C、D四组,每组18只,分别接受由Ⅰ组提供的经深低温保存1天、10天、1个月、3个月的肌腱。术后3天时、10天时分别从A组、B组、C组、D组随机抽取3只,近端自肌肉肌腱移行处,远端自跟腱附着处切断(包括近、远端吻合口),进行大体观察,组织学观察;术后4W、8W时分别从A组、B组、C组、D组随机抽取6只,进行大体观察,组织学观察,最大载荷测定。结果:保存10天以上的三个时间段的肌腱移植后和保存1天的比较,保存10天以上的肌腱术后粘连较轻,肌腱抗拉力强度降低。保存10天以上三个时间段的肌腱移植后大体观察、组织学观察无明显差异;最大载荷测定,结果:分析无统计学意义(P>0.05)。结论:肌腱在深低温(-80℃)环境中长时间保存后,其用于功能修复,愈合情况无明显差别。 Objective:Discusses the best time of deep low temperature (-80℃) to preserve the allogenic tendon. Methods:Body weight 250-300g female and male does not divide SD rats 144, divide into stochasticallyⅠ, Ⅱ Two groups. There are 72 rats in Each group. The groupⅠ supplies the tendons, provides144 the tendon which uses for the transplant. The groupⅡ also is stochastic divides into A, B, C, the D four groups, There are 18 in each group of, accept the tendons separately by the groupⅠ provides preserves for 1 day, 10 day, 1 month, 3 month after the deep low temperature. The operation latter 1st day and 10th day ,takes tendons from group A, group B, group C and group D, proximate from muscle carryover to tendon, far end from where achilles tendon adhesion to (including the Stomas of proximate and far end), to observe them in general, then to observe in histology; The operation latter 4th week and 8th week. takes suction 6 tendons stochastical from group A , group B, group C and group D, to observe them in general and sections histology; then to survey maximum load. Results:The experimental result indicate that the post transplant tendons preserve three time segmental of above 10 days to compare to what preserve 1 day, the tendons that preserve above 10days that Adhesions is gentler than what preserve 1 day, and the tensile resistance strength of the tendons decreases. There are not obvious difference in the post transplant tendons that preserve three time segmental of above 10 days in general observation and histology observe, to survey maximum load of the tendons, and then to analyze with statistics, there are not difference. Conclusions : The experiment to prove that the tendons can preserve a long time in deep low temperature(-80℃), and then use in function restoration, the healing information is not obvious difference.
