摘要
目的 探讨IL 1β在实验性视网膜脱离及复位状态下的表达改变及在实验性视网膜脱离中可能起到的作用。方法 应用HealonGV (14 g·L-1透明脂酸钠 )注入SD大鼠的视网膜下。放射免疫法检测脱离后不同时间点及复位后视网膜神经上皮的IL 1β蛋白含量。通过免疫组化定位IL 1β的表达。应用图像分析系统分析脱离区色素上皮、未脱离区及复位区色素上皮IL 1β表达的差异。 结果 在正常情况下 ,视网膜不表达IL 1β。视网膜脱离后表达于Muller细胞、胶质细胞、血管内皮细胞及色素上皮细胞的胞浆。IL 1β的表达在视网膜神经上皮于脱离后的第 7天达到高峰 ,然后下降。只要视网膜脱离存在 ,IL 1β的表达将维持在较低的水平上。视网膜复位后 ,视网膜神经上皮IL 1β的表达比未复位组下降。在色素上皮 ,IL 1β在脱离区的表达明显高于未脱离区及复位区。 结论 视网膜神经上皮与色素上皮之间的脱离导致视网膜表达IL 1β ,视网膜复位时表达下降。IL
Objective To investigate the change of expression of IL-1β in detached and reattached retina and the possible role of IL-1β in experimental retinal detachment.Methods Healon GV (14g·L -1 hyaluronate) were injected right into the subretinal space of SD rats. Retinas from the SD rats were detected at different period and the level of IL-1β in detached and reattached retina were detected by radio-immune method. Wax-embedded sections were labeled with the IL-1β antibody to determine the location of IL-1β. The difference of IL-1β expression in normal, detached and reattached retinal pigment epithelial cells (RPE) was analyzed by image analysis system.Results IL-1β was not labeled in normal retina.After experimental retinal detachment, Muller cells, glia cells,endothelial cells, RPE cells were labeled with IL-1β antibody. And the level of IL-1β in neuroretina was peaked at 7 days after retinal detachment, and remained at low level as long as the retina detached. The level of IL-1β in reattached retina was lower than that in detached retina. The expression of IL-1β in RPE of detached area was stronger than that in attached and reattached area.Conclusion The expression of IL-1β is result from the separation of neuroretina and RPE. Reattachment could lower the expression of IL-1β in retina.IL-1β is a key factor in retinal detachment.
