摘要
目的 对比研究两性霉素B与维生素E对大鼠晶状体钠泵α亚单位mRNA表达的影响。方法 将两性霉素B与维生素E注射到大鼠体内 ,连续给药 10d ,并设置生理盐水对照。手术取出晶状体 ,用逆转录聚合酶链式反应法测定各组大鼠晶状体钠泵α亚单位在mRNA水平的表达状况。结果 正常大鼠晶状体钠泵α重整异构体在mRNA水平的表达以α2 为主 ,与α1及α3 比较差异有显著性意义 (P <0 .0 1)。两性霉素作用后的晶状体钠泵α亚单位在mRNA水平的表达明显升高 ,尤其是α1和α3 的升高具有显著性意义 (P <0 .0 1)。维生素E作用后的晶状体钠泵α亚单位仅α1重整异构体在mRNA水平的表达明显增高 (P <0 .0 1)。结论 两性霉素B与维生素E通过作用于大鼠晶状体钠泵α亚单位不同的重整异构体 ,提高其在mRNA水平的表达。
Objective To investigate the influence of amphotericin B and vitamin E on expression of mRNA of rat lens sodium pump alpha subunit.Methods Amphotericin B and vitamin E were injected into the rats for 10 days.We took out the rat lens,and the expression of mRNA of rat lens sodium pump alpha subunit were examinated by reverse transcription polymerase chain reaction (RT-PCR).Results In normal rat lens,the expression of mRNA of sodium pump alpha subunit were dominated by α 2,the comparison among the three groups (α 1,α 2,α 3) showed significant difference(P<0.01).While amphotericin B acting on the rat lens , the expression of mRNA of sodium pump alpha subunit were enhanced significantly, especially demonstrated in α 1 and α 3(P<0.01). While vitamin E acting on the rat lens,the expression of mRNA of sodium pump alpha subunit were enhanced only in α 1 (P<0.01).Conclusion Amphotericin B and vitamin E could enhance the expression of mRNA of sodium pump alpha subunit owing to affection on the different α-isoforms.
