氧化铝脱色-中压柱层析纯化茄尼醇的工艺 被引量:4

摘要 为了获得高纯度茄尼醇,开发了一种氧化铝脱色-中压柱层析纯化废次烟叶中茄尼醇粗品的方法。确定了氧化铝吸附脱色的工艺条件:6号溶剂油作为溶剂,吸附时间40min,茄尼醇粗品与氧化铝的质量比为1∶8,洗脱剂(6号溶剂油)与茄尼醇粗品的比例为100∶1(mL/g),氧化铝吸附脱色后,茄尼醇收率92.8%,纯度为68.32%;采用连续中压柱层析法对茄尼醇进行了纯化,优化的连续中压柱层析条件为:以300~400目层析用中性氧化铝为填料,6号溶剂油-丙酮为洗脱剂(30∶1),柱使用压力为1.0MPa,柱层析后茄尼醇收率78.6%,纯度为94.37%。结果表明,该文建立的纯化工艺适用于茄尼醇的生产。 In order to obtain high purity of solanesol,an alumina decoloration-medium pressure column chromatography method was developed for the purification of solanesol in the crude extract from waste tobacco leaves.The optimum technology conditions of alumina adsorption decoloration were determined as follows:6 # solvent naphtha was used as solvent,adsorption time was 40 min,ratio of crude extract of solanesol to alumina was 1︰8,ratio of eluant(6#solvent naphtha)to crude extract of solanesol was 100︰1(mL/g).The results showed that under the optimal decoloration conditions of alumina,the yield of solanesol was 92.8%and the purity of solanesol was 68.32%.After decoloration by alumina,solanesol was purified by medium-pressure column chromatography.The optimum conditions of medium-pressure column chromatography were determined as follows:300~400 screen mesh of neutral alumina was used as stuffing,6#naphtha to acetone(30︰1)was used as eluant and the column pressure was 1.0 MPa.Under the optimized medium-pressure column chromatography conditions,the yield of solanesol was 78.6%and the purity of solanesol was 94.37%.It can be demonstrated that this is a good purification technique for the production of solanesol.
