摘要
目的 :探讨p38信号通路 (p38MAPK)在白细胞介素 1 (IL 1 ) β上调肾小球系膜细胞表达白细胞介素 6 (IL 6 )中的作用。方法 :应用WesternBlotting检测p38MAPK在IL 1 β诱导的肾小球系膜细胞炎症反应中的活化程度 ,应用逆转录 聚合酶链反应 (RT PCR)和ELISA法检测IL 1 β诱导的系膜细胞促炎症介质IL 6的表达水平并观察p38MAPK特异性抑制剂SB2 0 35 80对其mRNA的转录和蛋白质生成的影响。结果 :IL 1 β以时间和剂量依赖方式刺激系膜细胞引起p38MAPK的活化 ,并明显上调系膜细胞IL 6表达。SB2 0 35 80以剂量依赖方式从基因转录和翻译水平显著抑制IL 1 β诱导的IL 6表达。 结论 :p38MAPK在IL 1 β诱导的肾小球系膜细胞炎症反应产生炎症介质IL
Objective:To investigate the action and mechanism of p38 mitogen-activated protein kinase(p38MAPK) in up-regulating interleukin 6 expression in rat glomerular mesangial cells induced by interleukin 1β.Methods:Activated p38MAPK were detected with Western Blotting, while the expression of interleukin 6 were measured with RT-PCR and ELISA,and investigated the effect of SB203580 on transcription of interleukin 6 mRNA and translation of protein in rat mesangial cells stimulated by interleukin 1β.Results:Interleukin 1β can activate p38MAPK high expression in time-dependent and dosage-dependent manner, and up-regulated interleukin 6 expression in rat glomerular mesangial cells. SB203580, a p38MAPK inhibitor,inhibited not only the transcription of interleukin 6 mRNA but also the translation of protein up-regulated by interleukin 1β in dosage-dependent manner. Conclusion:p38MAPK may play an important role in high expression of proinflammatory factor-interleukin 6 in rat glomerular mesangial cells induced by interleukin 1β.
