摘要
目的 :探讨反义c myc寡核苷酸 (c mycASODN)对半乳糖诱导的兔晶状体上皮细胞 (LEC)中c Myc蛋白表达及细胞周期的影响 .方法 :人工合成与c myc基因第二外显子翻译起始区序列互补的寡核苷酸并磷酸化修饰 ,用半乳糖和c mycASODN处理体外培养的兔晶状体 ,流式细胞术检测c mycA SODN对半乳糖诱导的LEC中c Myc蛋白表达水平及细胞周期的影响 .结果 :2 .5~ 1 5 .0 μmol/Lc mycASODN能下调半乳糖诱导的晶状体上皮细胞中c Myc蛋白的表达 ,荧光强度从2 1 4 .7± 0 .6降至 90 .5± 8.4 ,其作用有剂量依赖性 (r=- 0 .983,P <0 .0 1 ) ,同时可降低S期细胞数 ,从 5 .5 4降至 4 .5 9.结论 :c mycASODN能抑制半乳糖诱导的晶体上皮细胞中c Myc蛋白的表达水平 ;阻止细胞进入S期 。
AIM: To study the effects of antisense oligodeoxynucleotide c myc ( c myc ASODN) on the expression of c Myc protein and the cell cycle of rabbit lens epithelial cells (LEC) induced by galactose. METHODS: From translation initiation region of second extron of c myc gene, oligonucleotides were synthesized and modified with phosphorothioate. Rabbit lens were treated with galactose and c myc ASODN and the effects of c myc ASODN on the level of c Myc protein expression and the cell cycle of LEC induced by galactose were detected by flow cytometry. RESULTS: A 2.5- 15.0 μmol/L of c myc ASODN reduced the level of c Myc protein expression on LEC induced by galactose in a dose dependent manner, counts decreased from 214.7± 0.6 to 90.5 ± 8.4 ( r =-0.983, P <0.01), and the number of S phase cell of LEC decreased from 5.54 to 4.59. CONCLUSION: c Myc ASODN can inhibit the level of c Myc protein expression on LEC induced by galactose, check the entry of cells into the S phase and inhibit the proliferation of LEC.
