肉豆蔻乙醇提取物对D-氨基半乳糖中毒大鼠急性肝损伤的保护作用 被引量:6

摘要 [目的]探讨肉豆蔻乙醇提取物对D-氨基半乳糖中毒大鼠急性肝损伤的保护作用。[方法]用腹D-氨基半乳糖中毒制作大鼠急性肝损伤模型,实验大鼠随机分为正常对照组、肝损伤组、肉豆蔻低剂量组、肉豆蔻中剂量组和肉豆蔻高剂量组,分别灌胃给药6d。第5d除正常组腹腔注射等量的生理盐水之外,其他各组均一次性腹腔注射D-氨基半乳糖500mg/kg,造成急性肝损伤模型,36h后取血及肝组织,分别测定血清丙氨酸转氨酶(ALT)、天门冬氨酸氨基转氨酶(AST)、超氧化物歧化酶(SOD)活性,脂质过氧化物丙二醛(MDA)含量及肝组织超氧化物歧化酶活性、脂质过氧化物丙二醛含量,并作肝组织病理形态学检查。[结果]肉豆蔻中剂量组、高剂量组大鼠血清丙氨酸转氨酶、天门冬氨酸氨基转氨酶活性,血清及肝组织脂质过氧化物丙二醛含量明显低于损伤组,而超氧化物歧化酶活性明显高于损伤组。病理分级结果,肉豆蔻高剂量组变性及坏死较损伤组显著减轻。[结论]肉豆蔻乙醇提取物对D-氨基半乳糖中毒大鼠急性肝损伤具有呈量一效关系的保护作用。 To investigate the protective influence ofthe Nutmeg ethanol extract (NTEE) on D - galac tosamine intoxication induced acute liver lesion in rats. METHODS : The experimental models of acute liver injury were made by intraperitoneal injection of D - galactosamine intoxication,50 rats were randomly divided into (A)normal group,(B)injury group, (C) NTEE low dosage group,(D) NTEE middle dosage group (E) NTEE high dosage group, and the NTEE was perfused to stomach for six days.In the five days,the 0.09 salt solution was injected into peritoneum in nonnal group, but the D - galactosamine intoxication 500 mg/kg was injected into peritoneum in the rest groups one time to induce the models of acute liver lesion. thirty - six h later, the blood and liver tissue were taken out and itwas respectively measured that the Alanine aminotransferase( ALT)、Asparate transferasr( AST) 、Superoxide dismutase activities ( SOD ) 、 Lipid peroxide malondialdehyde(MDA) contents of serum and superoxide dis mutase superoxide dis mutase activities, lipid peroxide malondialdehyde contents and protein contents of liver tissue were measured, At the sametime, Pathological morphologies of hepatic tissue were observed. RESULTS: In the middle and greater dosage of Nutmeg group alanine aminotransferase, asparate transferase activities, lipid peroxide malondialdehyde contents of serum and liver tissue were much lower than those in injury group; While superoxide dismutase activities in serum and hepatic tissue were much higher than those in injury group,The pathological grading result was that the greater dosage of NTEE group compared with injury group were markedly difference in deterioration and necrosis. CONCLUSION: Nutmeg ethanol extract has protective influence on acute liver injured by D - galactosamine intoxication in rast on a dose - dependent manner.
