应用AFLP-DNA指纹技术鉴定梅花品种的研究 被引量:8

摘要 应用银染法AFLP技术,通过筛选的7对Mse I-EcoR I引物组合,建立了162个梅花品种的175个样品指纹。使用NTSYSpc2.1t软件,计算遗传相似性,以SAHN Clustering进行不加权成对算术平均法(UPGMA)聚类分析。其中引物组合E-ACT/M-CAG和E-GA/M-CTC分别仅有两个样品不能区分开,其余引物组合均能鉴别175个样品中的164个以上的样品。7对引物组合的平均鉴别效率达到95.89%,两对引物组合可以鉴别所有样品。以引物组合E-GA/M-CTC为例进行了品种鉴定的聚类分析。所以,用AFLP-DNA指纹图谱来鉴别梅花品种资源,效率是很高的。 The silver-stained AFLP analysis was employed with Mse I-EcoR I digest and primers,finger- prints of the 179 samples of 162 cultivars were obtained.With 7 combinations of primers,NTSYSpc 2.It was used to SAHN Clustering of UPGMA and cluster analysis was done.The primer combinations E-ACT/M-CAG and E-GA/M-CTC did not distinguished two cultivars among 162 cultivars.The 7 combinations of primers had an average identifying percentage of 95.89 % and all of the cultivars could be distinguished with two primers.The primer E-GA/M-CTC was taken as an example to conduct cluster analysis by using NTSYS software to reflect the similarity among varieties.Nearly all results indicated that AFLP is highly effective in identifying Mei cultivars.
