摘要
胶质瘤是成人颅内最常见的恶性肿瘤。本研究探讨人胶质瘤细胞SWO-38两亚株(SWOZ1和SWOZ2)异质性的蛋白质分子机制。方法:采用Western blot法检测SWO-38及其两亚株中鼠双微粒体基因-2(Murine double minute-2,MDM2)的表达情况;应用免疫共沉淀技术检测两亚株中MDM2相关蛋白的表达情况。结果:在SWO-38及其两亚株中均能检测到MDM2的表达,在其免疫共沉淀物中,Mr 37000处有一蛋白条带在SWOZ2中比在SWOZ1中明显深染。结论:人胶质瘤细胞SWO-38的两亚株,在MDM2传导通路中于Mr约37000处的某蛋白表达量不同,而此差异蛋白可能与胶质瘤异质性的产生密切相关。
BACKGROUND & OBJECTIVE: Glioma is the most common brain tumor in adult. This study was undertaken to investigate the protein molecular mechanism of tumor heterogeneity of gliomas with two sublines(SWOZ1 and SWOZ2) of SWO-38.METHODS: MDM2 expression in SWO-38 and its two sublines were determined by using Western blot analysis. Then, co-im-munoprecipitation method was used to exam the expression of MDM2 binding proteins in two sublines. RESULTS: Expression of MDM2 was observed in all SWO-38 and its two sublines, and there was a major difference between two sublines in the MDM2' S immuno-precipitation compond which was in the protein molecule weight of 37000. CONCLUSION: The difference in the MDM2 transmit channel protein with molecule weight of 37000 between two sublines of SWO -38 may associated with heterogeneity of the gliomas.
