蛋白酶抑制剂对肝硬化大鼠肝移植围手术期的影响 被引量:3

摘要 目的 探讨蛋白酶抑制剂在肝硬化大鼠肝移植围手术期的保护作用。方法 采用二袖套法大鼠原位肝移植动物模型。 6 4只SD大鼠随机均分成两组 :实验组 (A组 ) ,于受体手术前 30min自静脉注射乌司他丁 (2× 10 4U L) 1.5ml kg ;对照组 (B组 ) ,于受体手术前 30min自静脉注射同等体积生理盐水。每组受体的一半 (n =8)用于观察存活率 ,另一半 (n =8)用于移植肝脏再灌注 2h后取血及肝脏检测。结果 实验组的 1W存活率、胆汁分泌量、抗氧化酶活力均明显高于对照组 (P <0 .0 5 ) ,血清ALT、AST、LDH、肝组织中髓过氧化物酶 (MPO)含量及过氧化产物含量均明显低于对照组 (P <0 .0 5 )。结论 蛋白酶抑制剂乌司他丁能够保护移植肝脏功能 ,对行原位肝移植的肝硬化大鼠有较好的抗手术侵袭作用。 Objective To investigate the protective effect of protease inhibitor in perioperation of orthotopic liver transplantation in hepatocirrhosis rats. Methods The model was established with two- cuff technique. Sixty-four SD rats were randomly divided into 2 groups (n=32). Group A: Before operation of recipients, Ulinastatin was administered to animals. Group B: Before operation of recipients, the same volume of brine was administered to animals. One half of each group were used to investigate the week survival rate of recipients, and another half of each group were used to take sample of blood and hepatic tissue after 2 hours of reperfusion. Results The 1 week survival rate, content of bile and antioxidase activity was higher in group A than that in group B (P<0.05). Meanwhile, serum ALT, AST, L, DH, MPO and superoxide in hepatic tissue was lower in group A than that in group B (P<0.05). Conclusions Ulinastatin, a protease inhibitor, could protect the function of grafts during liver transplantation and have good impact in antagonism operative foray for hepatocirrhosis rat undergone orthotopic liver transplantation.
