川芎嗪和参麦注射液对老龄大鼠脑缺血/再灌注前列腺素和β-内啡肽的影响 被引量:5

Effect of Ligustrazine and Shenmai Injection on Prostacycline and β-Endorphin Content in Aged Rats with Cerebral Ischemia/Reperfusion

摘要 目的:从血栓素 A_2(TXA_2)与前列环素(PGI_2)平衡失调和β-内啡肽(β-EP)的变化方面研究川芎嗪和参麦注射液及川芎嗪加参麦注射液对老龄大鼠脑缺血/再灌注损伤防护作用的机制,为防治脑梗塞提供实验依据。方法:老龄(20月龄以上)大鼠分为老龄对照组、老龄模型组、尼莫通组、川芎嗪组、参麦组、川芎嗪加参麦组(川芎参麦组),观察大鼠全脑缺血30 min 再灌注60 min 后血浆血栓素 B_2(TXB_2)、6-酮-前列腺素F_(1α)(6-keto-PGF_(1α))、β-EP 含量和脑组织 TXB_2和6-keto-PGF_(1α)的水平。结果:老龄模型组血浆β-EP 水平和脑组织 TXB_2/6-keto-PGF_(1α)高于老龄对照组(P<0.05);尼莫通组β-EP 低于老龄模型组(P<0.05),接近老龄对照组;尼莫通组和川芎参麦组及参麦组脑组织 TXB_2/6-keto-PGF_(1α)低于老龄模型组(P<0.05,P<0.01),接近老龄对照组。结论:脑组织中以 TXA_2为优势的 TXA_2/PGI_2平衡失调和β-EP含量增高与老龄大鼠脑缺血/再灌注脑组织损伤有关。尼莫通和参麦注射液及川芎嗪与参麦注射液配伍均能调节 TXA_2与 PGI_2平衡失调,这可能是其拮抗脑组织损伤的机制之一。 Objective:To study the protective mechanism of ligustrazine (LT),Shenmai Injection (SI) and the combination of the above-men- tioned injections (LSI) on cerebral ischemia/reperfusion injury by means of observing the imbalance of thromboxane A_2 (TXA_2) and prostacyclin (PGI_2),and the changes of β-endorphin level (β-EP) in the aged rats with cerebral ischemia/reperfusion injury,so as to provided the scientific basis for the prevention and treatment of cerebral infarction.Methods:The aged rats (20 montha or more) were divided into six groups:normal control group,model group,nimotop group,LT group,SI group and LSI group.The plasma TXB_2,6-ke- to-PGF_(1α) and β-EP level and the brain tissue TXB_2,6-keto-PGF_(1α) level were observed in the rats with 60 rain reperfusion after 30 min cerebral ischemia.Results:The plasma β-EP level and the brain tissue TXB_2/6-keto-PG_(1α) ratio in the aged model group were higher than those in the aged control group (P<0.05).The β-EP content in nimotop group was lower than that in the aged model group,and close to that in aged control group (P<0.05).The TXB_2/6-keto-PGF_(1α) ratio in the nimotop group,LSI group and SI group was lower than that in aged model group (P<0.05,P<0.01),and close to that in aged control group (P<0.05).Conclusion:The imbalance of brain tissue TXA_2/PGI_2 and the increase of β-EP content were correlated with the cerebral ischemia/reperfusion injury in aged rats.The nimotop,SI and LSI all could adjust the imbalance of TXA_2-PGI_2,which might be one of the mechanisms that protect against brain is- chemia/reperfusion injury.
