回春口服液对老年大鼠弓状核神经元影响的光、电镜定量研究

摘要 目的:探讨中药的抗衰老作用和开发抗衰老药品。方法:40只老年 SD 大鼠随机分为老年组和回春口服液组,20只成年 SD 大鼠为对照组,采用光、电镜技术和体视学方法对回春口服液延缓弓状核神经元衰老作用进行了形态定量研究。结果:3组间弓状核神经元的面积分数、面数密度、胞体和胞核的等效直径无显著变化(P>0.05);老年组弓状核暗神经元内线粒体和溶酶体的体积分数和面数分数明显增加(P<0.05),粗面内质网、高尔基复合体、游离核糖体和分泌颗粒的体积分数和面数密度显著下降(P<0.05);回春口服液组与对照组之间弓状核暗神经元内细胞器和分泌颗粒的体积分数和面数密度差异无显著性(P>0.05)。结论:回春口服液具有延缓弓状核神经元衰老的作用。 Objective:To investigate anti-aging effect of Huichun Oral Liquid (HCOL) for developing new anti-aging drugs.Methods:Forty aged Sprage-Dawley rats were divided into aged group and HCOL treated group,and 20 healthy adult SD rats were taken as control. The effect of HCOL on morphology of arcuate nucleus neurons was observed by light and electron microscope.Results:Area fraction (AF) and numerical density (ND) on area of arcuate neurons and equivalent diameter of neuronal soma and nucleus had no significant change in all of the three groups.In the aged group,the volumetric fraction (VF) and ND on area of mitochondria and lysosome of dark neurons in arcuate nucleus were increased (P<0.05),while the AF and ND on area of the rough endoplasmic retieulum,Golgi com- plex,free ribosome and secretory granule were decreased (P<0.05).The organelles and secretory granules of dark neurons had no sig- nificant difference between the control and the HCOL group.Conclusion:The neuroendocrine function disorder occurred in senility might be correlated with the aging of arcuate nucleus neuron and HCOL could retard the aging process.
