补肾活血方对脑缺血小鼠智能及大脑皮层胆碱能系统的影响 被引量:4

摘要 目的:观察补肾活血方对脑缺血小鼠学习记忆能力及大脑皮层胆碱乙酰化转移酶(ChAT)、胆碱能 M 受体(MR)和 N 受体(NR)活性的影响。方法:用双侧颈总动脉结扎缺血再灌术,建立小鼠脑缺血智能障碍模型。分别灌胃生理盐水(假手术组、缺血模型组)、补肾活血方(防治组、治疗组)和尼莫地平(阳性对照组),观察了小鼠被动回避反应学习记忆能力、大脑皮层 ChAT、MR 和 NR 活性的变化。结果:与缺血模型组比较,补肾活血方能显著提高模型小鼠学习记忆能力(P<0.01),明显提高大脑皮层中的 ChAT、MR、NR 活性(P<0.05,P<0.01),与尼莫地平作用相近。结论:补肾活血方有改善脑缺血小鼠学习记忆能力、增加小鼠大脑皮层 ChAT、MR 和 NR 活性的作用,对脑缺血有保护作用。 Objective:To observe the effects of Bushen Huoxue Recipe (BSHXR) on the learning and memory capability,choline acetyltrans- ferase (CHAT) of cerebral cortex,and the activity of cholinergic M and N receptors (MR,NR) in mice with cerebral ischemia.Meth- ods:The models of intelligence disturbance of mice with cerebral ischemia were established by the method of ischemia-reperfusion injury caused by the ligation of bilateral carotid arteries.The learning and memory capability of mice were detected by the experiment of evad- ing reaction.The model mice were randomly divided into 5 groups,i.e.sham operation group,ischemic model group (normal saline), BSHXR prevention-treated group,BSHXR treated group and positive control group (nimodine).The learning and memory capability, the ChAT of cerebral cortex,and the activity of cholinergic MR and NR of the mice were determined.Results:Compared with model group,BSHXR could obviously improve the learning and memory capability in model mice (P<0.01),significantly increase the ChAT of cerebral cortex and the activity of cholinergic MR and NR,which was similar to the effect of nimodine.Conclusion:BSHXR could improve the learning and memory capability,increase the ChAT of cerebral cortex and activity of cholinergic MR and NR of the mice with cerebral ischemia.Therefore,BSHXR has protective effect on cerebral ischemia in mice.
