复方虎桃合剂对大鼠肺纤维化形成影响的研究

摘要 目的:研究大鼠肺纤维化(PF)的病理过程及复方虎桃合剂(HT)对其治疗作用。方法:应用电子显微镜,网状纤维染色和 ELISA 方法检测大鼠肺纤维化的病理过程和胶原含量的改变。结果:模型组肺泡I型上皮细胞数目减少,Ⅱ型细胞增多且处于活化状态,含有大量活性物质的板层小体被释放到肺泡腔,大部分肺泡腔和小支气管塌陷,白细胞数目增多,同时血管内皮细胞损伤收缩;Ⅰ、Ⅲ型胶原含量逐渐升高。HT治疗组上述病理变化明显轻于模型组,Ⅰ、Ⅲ型胶原含量降低。结论:HT 合剂可能通过保护血管内皮细胞和肺泡上皮细胞以及抑制胶原生物合成起到对肺纤维化的防治作用。 Objective:To investigate the pathogenesis of pulmonary fibrosis and the effect of composite Hutao Mixture (CHTM) in treating pulmonary fibrosis in rats.Methods:Electron microscopy,reticulum fiber stain and ELISA test were used to observe the pathologic pro- cess of pulmonary fibrosis and to determine the change of collagen content.Results:In the model group,the number of type Ⅰ pneumo- cytes decreased and atrophied while type Ⅱ pneumocytes activated and hypertrophied,lamella bodies which contained large amount of active substance,was released into alveoli.Majority of pulmonary alveoli and small bronchi collapsed with increased white blood cells. The lumen of alveolar capillary contracted and narrowed in the same time as well as the content of collagen type Ⅰ and Ⅲ increased gradually.In the CHTM treated group the above-mentioned pathological changes were lighter significantly than those in the model group,and the content of collagen type Ⅰ and Ⅲ was less than that in the model group.Conclusion:CHTM could through protect the vascular endothelial cells and alveoli epithelial ceils to prevent and treat pulmonary fibrosis.
