摘要
目的:探讨促进肝化脾增量及缩短其再生周期的新途径。方法:应用三七总皂甙于自体肝细胞脾内移植(IHAT)的动物模型上,分别于移植术后2周对病理组织学、电镜形态、肝化脾匀浆谷丙转氨酶(ALT)含量及^(99m)Tc-HIDA 摄取试验进行观察分析。结果:三七总皂甙组(A 组)脾内肝细胞水肿、变性程度较轻,肝化脾匀浆 ALT 含量达(928.0±268.1)u/g,明显高于对照组(P<0.05),且^(99m)Tc-HIDA 摄取试验时 A组离体肝化脾显影较清晰,其放射性计数明显高于对照组(B 组,P<0.05)。结论:三七总皂甙在移植早期对脾内肝细胞有一定的抗损伤保护作用,从而促进了肝化脾的增量。
Objective:To find the new route for promoting the weight of liver tissue mass in spleen and to shorten the regeneration cycle of transplanted hepatocytes by means of protection against ischemia/reperfusion injury.Methods:Panax notoginsen saponins (PNGS) was administered to intrasplenic hepatocellular autotransplantation (IHAT) model with 70% partial hepatectomy.Histological examination was carried out under both light and electron microscope,and alanine transaminase(ALT)content in hepatized spleen homogenate deter- mination as well as ^(99m)Tc-HIDA intake test were assayed 2 weeks after transplantation.Results:The extent of hepatocellular degenera- tion appeared to be slighter in group A(IHAT+PNGS)than that in group B(IHAT alone),ALT content and radioactivity accumulation of ^(99m)Tc-HIDA were significantly higher in group A than those in group B.Conclusion:PNGS has certain effects in protecting the transplanted hepatocytes from ischemic/reperfusion injury in the early stage of transplantation,thus promoting the process of spleen hepatization.
