中药复方对肝纤维化小鼠2′,5′-寡腺苷酸合成酶活性影响和超微结构研究

Study of Chinese Compound Recipe on Immunologic Function and Ultrastructural Change in Preventing and Treating Liver Fibrosis in Mice

目的:研究中药复方(二参泽术汤和丹参桃芎汤)防治肝纤维化时,对机体免疫功能的影响和肝细胞超微结构的改变。方法:制备小鼠肝纤维化模型;观察其病理组织学和超微结构,并测定2′,5′-寡腺苷酸合成酶活性。结果:模型组肝细胞广泛脂肪样变性,汇管区大量胶原纤维增生。2′,5′-寡腺苷酸合成酶水平低下。二参泽术汤预防组肝组织超微结构基本正常,未见有胶原纤维增生;治疗组肝细胞间有少量胶原纤维增生。该药预防组和治疗组2′,5′-寡腺苷酸合成酶活性显著高于模型组(P 均<0.01)。丹参桃芎汤预防组和治疗组有严重的细胞变性坏死,胶原纤维大量增生,2′,5′-寡腺苷酸合成酶活性与对照组比较,有明显降低(P<0.01)。结论:在肝纤维化治疗时,机体免疫功能的高低与疗效有密切关系;二参泽术汤对提高肝纤维化小鼠机体的免疫功能和防治肝纤维化的效果均优于用丹参桃芎汤。2′,5′-寡腺苷酸合成酶的测定能反映肝纤维化的转归和预后。

Objective:To study the effect of traditional Chinese medicine on the immunologic function and ultrastructural change of liver cells in preventing and treating liver fibrosis.Methods:The mouse model of liver fibrosis was established,the pathohistology and ultrastruc- ture of liver cells was observed,and the dynamic determination of the 2′,5′-oligoadenylate synthetase activity (OASA) was carried out. Results:In the model group,widespread fatty degeneration in liver cells,large amount of collagenous fiber was found in the portal area, and OASA lowered in comparing with the normal group.In Ershen Zezhu decoction(ESZZ) prevention group,the liver ultrastructure was normal,and no collagenous fiber was found in the liver tissue.And in the ESZZ treatment group,only small amount of collagenous fiber among the liver cells was seen.The OASA in the prevention and treatment group of ESZZ increased obviously compared with that of the model group (prevention group P<0.01,treatment group P<0.01).However,in the prevention and treatment group of Dan- shen Taoxiong decoction(DSTX),the degeneration and necrosis of liver cells were severe,the collagenous fiber proliferated markedly, and the OASA reduced.The results revealed that the pathologic changes of the liver tissue was consistent with the changes of immuno- logic function in experiment group.Conclusion:ESZZ was better than DSTX in enhancing the immunologic function of liver fibrosis in mice and in preventing and treating liver fibrosis.Experimental study showed that the immunological function is closely related with the therapeutical effect during the treatment of liver fibrosis,and the determination of OASA could reflect the prognosis of liver fibrosis, which is important clinically.
