摘要
目的:观察中药钙通道阻滞剂川芎嗪(TMP)对 HL60/HT 耐药细胞多种化疗药物的增敏作用。方法:采用四氮唑蓝(MTT)法细胞毒性实验作为药物敏感实验,用流式细胞仪测量细胞内药物浓度。结果:TMP 对多药耐药 HL60/HT 细胞系具有抑制生长作用并表现出剂量依赖关系,32、100μg/ml 非细胞毒剂量的 TMP 和多种化疗药物联合应用可以提高化疗药物的细胞毒性作用,逆转倍数为1.25~8.0,相对逆转效率为21.42%~87.90%。流式细胞仪测量细胞内柔红霉素(DNR)浓度发现,TMP 可以增加 HL60/HT 细胞内 DNR 的浓度。结论:TMP 可能是一个有潜力的中药白血病化疗药物增敏剂。
To investigate the enhancing effect of chemotherapy sensitivity of tetramethylpyrazine(TMP),a Chinese drug of calci- um channel blocker,on multi-drug resistant(MDR)leukemia cell line HL60/HT.Methods:The drug sensitivity test was performed with methyl thiozolyl tetrazolium(MTT)cytotoxic test,and the intracellular drug concentration was measured by flow cytometry.Re- sults:TMP could inhibit the growth of the MDR HL60/HT cell line with a dose-dependent manner,and the non-cytotoxical TMP(32 and 100 μg/ml)combined with multiple chemotherapeutic agents could enhance the cytotoxicity of the latter to HL60/HT cell line. The effectiveness could be enhanced by 1.25-8.0 times with a relative reversal efficiency rate reaching 21.42% to 87.90%.Intracel- lular concentration of daunorubicin(DNR)measurement by flow cytometry showed that TMP could elevate the concentration of DNR in HL60/HT cells.Conclusion:TMP could be a potent chemotherapy sensitivity enhancing agent for leukemia.
