摘要
目的 探讨人白细胞介素2(IL-2)基因转移对卵巢癌细胞周期和转移生长因子β_1(TGF-β_1)表达的影响。方法 采用流式细胞术探讨IL-2基因转移对卵巢癌细胞SKOV3细胞周期及TGF-β_1表达的影响。结果 SKOV3/IL-2细胞GO、G1期细胞比例明显高于对照组,而S、M期细胞明显少于对照组,P<0.001,表现为G0/G1期阻滞。SKOV3、SKOV3/Neo、SKOV3/IL-2细胞TGF-β_1阳性百分率及荧光强度分别为98.64%、97.00%、99.52%和143、138、163,SKOV3/IL-2平均荧光强度较亲本细胞明显提高,P<0.001,差别非常显著。结论 IL-2基因转移可引起卵巢癌细胞系SKOV3细胞周期G0/G1期阻滞,TGF-β_1表达的增强有可能是IL-2基因转移诱发细胞增殖抑制的原因之一。
Objective To study the effect of IL-2 gene transfer on cell cycle and TGF-β1 expression of ovarian cancer cell line SKOV3. Methods IL-2 gene was introduced into ovarian cancer cell line SKOV3, cell cycle and TGF-β1 expression was studied by fluorocytometry. Results The cell cycle study showed that SKOV3/IL-2 cell was block to phase G1/G0. The TGF positive rate and mean fluorescence density of SKOV3, SKOV3/Neo and SKOV3IL-2 was 98. 64%, 97. 00%, 99. 52% and 143, 138, 163 respectively. There were significance difference between SKOV3, SKOV3/ Neo and SKOV3/IL-2 cells on mean fluorescence density, P< 0 . 001 . Conclusion IL-2 gene transfer can induce Gl/GO phase cell cycle blocking and upregulate TGF-β1 expression of SKOV3 cell line.
