肥厚性瘢痕TGF-β和胶原酶mRNA表达及定量研究

摘要 本文采用原位杂交、图像分析和SDS-聚丙烯酰胺凝胶加胶原底物电泳法观测了肥厚性瘢痕中TGF-β和胶原酶mRNA的表达及酶活性,结果发现在肥厚性瘢痕组织中TGF-β表达较正常皮肤明显增高,也比胶原酶的表达明显增加,平均积分光密度分别增加了12倍和8倍(P<0.001),TGF-β与胶原酶表达呈显著负相关,肥厚性瘢痕组织中胶原酶活性降低,提示TGF-β抑制胶原酶表达和活性,阻止胶原降解,导致肥厚性瘢痕的形成。 The Expression of mRNA for transforming growth factor β(TGF β) and collagenase(MMP 1) in hypertrophic scar (HTS) tissues were studied with in situ hybridization combined with image analysis, and the collagenase activity was investigated by electrophoresis on sodium dodecyl sulfate substrate gels. The results revealed that the expressiion of mRAN for TGF β increased in HTS compared with normal skin, and was also greatly higher than the expression of MMP 1. The average integral optical density of mRNA for TGF β in HTS increased 12-fold compared with that in normal skin (P<0 001) and 8 fold with MMP 1(P<0 001). Both TGF β and MMP 1 expression showed high negative correlation. The collagenase activity was reduced in HTS tissues. These findings suggested that TGF β inhibitied the expression and activity of MMP 1, and induced the formation of HTS.
