异搏定对离体大鼠心脏钙超载心肌损伤保护作用的研究

摘要 目的 探讨心肌钙通道阻滞剂异搏定(Verapamil,VP)对钙超载所致心肌损伤的作用。方法 采用在Lengend-off灌流装置上制作的缺钙-复钙心肌模型,对24例大鼠心脏进行离体灌流,检测心肌钙含量、灌流液中乳酸脱氢酶(LDH)及心肌蛋白漏出量。结果 以1mg/L的VP灌注心脏未能减轻心肌钙超载,损伤心肌的LDH及蛋白漏出量无明显减少(P>0.05),光镜下观察心肌损伤未减轻。结论 VP对钙反常心肌损伤无保护作用。异搏定对于治疗钙超载相关的心肌损伤价值不大。 Objective To explore the effect of the slow channel calcium antagonist verapamil(VP) in preventing calcium overload related heart injury. Methods 24 isolated rat hearts were perfused on Lengendoff perfusion equipment. The effect of VP upon the perfused heart models were determined by measuring calcium content of the myocardium,the LDH release,the protein loss and the coronary flow. Results The calcium content of the myocardium,the LDH release,the protein loss and the coronary flow had not been significantly reduced when the rat hearts were perfused by 1mg/L VP. Conclusion VP has no protective effect on the myocardium injury during calcium paradox. The massive influx of Ca2+ during calcium paradox is not through the slow calcium channel. VP may has little value in treating calcium overload related injury in clinic.
