摘要
采用无血清成年大鼠肺组织培养,液闪测3H-胆碱掺入PC量,现察hEGF对成年大鼠PS合成的影响。结果显示:①hEGF促进肺组织3H-胆碱掺入量,量-效关系显著。②10-9mol/LhEGF作用8h后,可显著增加3H-胆碱掺入量P<0.01),16h达高峰。③PKC抑制剂10-5mol/LH7可显著降低10-9mol/LhEGF组3H-胆碱掺入量(P<0.01)。④hEGF对肺组织LDH释放量无显著影响,提示本实验各处理因素对肺组织无细胞毒性。结果表明EGF可促进成年大鼠PS合成,PKC参与介导EGF促PS合成效应。
The effect of hUman epidennal growth factor on PS synthesis was studied in cultured ratlung explants without serum, and amount of 3H-choline incorporation into PC wasdetermined. The results showed that: rohEGF increased 3H-chollne inocrporation inadedependent rnarmer. ②10-9mol/L hEGF increased 3H-choline incomption into PC after sh(P < 0. 05). ③H7,a PKC ichbitor decreased 3H-choline incorporation induced by(P < 0. 01). ④the release of LDH from lung tissues could not be changed by hEGF. Theresults showed that EGF could enhance the adult PS synthesis mediated by PKC.
