兔膀胱5-氟尿嘧啶微球化学栓塞的实验研究

摘要 采用以5-氟尿嘧啶(5-Fu)、人血清白蛋白为材料制成的微球,注入实验兔髂内动脉,测定髂内动脉注入微球后的血药浓度及膀胱组织药物浓度。同时对5只兔以同法注入等量常规5-Fu作为对照。结果:制成的微球直径35~75μm,含药量均为10%。体外研究微球24h释药量为35%,实验组血药高峰浓度为23.8μg/ml,明显低于对照组(P<0.05).出现时间也比对照组晚30min.注药后4h兔膀胱组织中5-Fu的量则是33.6μg/g,显著高于对照组(P<0.01)。认为注入5-Fu微球于肿瘤区,发挥其局部化疗的作用,明显优于单纯药物动脉内注入法。5-Fu微球注入法是一种有希望的抗肿瘤疗法,为今后的研究方向提供了依据。 5-Fu concentrations in blood and bladder tissue were detectcd to study the preparation and pharmacological charactcristics of 5-Fu microsphere in 10 rabbits infused with 5-Fu microsphere and 5 rabbits (thc control group)infused in the same dosage via iliac artery. The results showed that the 5-Fu microsphere ranged from 35 to 75μm in diameter and the content of 5-Fu in 5-Fu microsphere is about 10%. The peak of 5-Fu concentration in serum of the experimental group was 23. 8μg/ml, much lower than that of the control (P < 0. 05). 5-Fu concentration in bladdet tissue of the experimental group was 33. 68μg/g. much higher than that of the control (P<0. 01). The experiment indicated that 5-Fu microsphere chemotherapy might be effective.
