骨形态发生蛋白的提纯及功能研究

摘要 1目的 提纯牛骨形态发生蛋白 (BMP) ,测定其分子质量和氨基酸组成 ,分析其功能。2方法  BMP经 Sephacryl S- 2 0 0柱层析纯化 ,SDS- PAGE测定其分子质量 ,氨基酸分析仪测定氨基酸组成。 3结果  BMP分子质量范围为 13.9~ 6 3.5 ku,主要由 13.9,18.4,2 5 .8,35 .7ku等 4种分子质量的组分组成 ;氨基酸分析表明 BMP主要由 17种氨基酸组成 ,其中以甘氨酸、脯氨酸、谷氨酸、丙氨酸等含量最高。4结论  BMP的 4种组分均由甘氨酸、脯氨酸、谷氨酸、丙氨酸等 17种氨基酸组成。部分纯化 BMP中 ,不溶性非胶原蛋白和非蛋白类物质对 Objective To prepare and purify bovine bone morphogenetic protein(BMP) ,measure its m olecular weight (MW) and amino acid components,and analyze its biological activity. Methods MBP was purified by sephacryl S-2 0 0 chromato graphy,Its MW was determined by SDS-PAGE and its amino acid components was analysed by amino acid analyzer. Results The MW range of BMP was from 13.9to 6 3.5 ku,BMP mainly contained four constituents with the MW 13.9,18.4,2 5 .8, 35 .7ku.BMP was com posed of17kinds of amino acids and the contents of glycine,proline,glutamic acid and alanine were the highest. Conclusion Purified BMP was composed of 17kinds of amino acids including glycine ,poline,glutamic acid,alanine, etc.Insoluble non-collagen proteins(i NCP) together with non-protein substance may have som e enhancing effect in the bone inductive activity of BMP.
