粉防己碱对血管平滑肌细胞增殖及癌基因表达的影响

EFFECTS OF TETRANDRINE ON PROLIFERATION OF VASCULAR SMOOTH MUSCLE CELLS AND EXPRESSION OF ONCOGENES

采用血管紧张素Ⅱ建立了培养的血管平滑肌细胞增殖模型。用H3-胸腺嘧啶核苷掺入法、流式细胞术、免疫细胞化学及Northernblot方法．观察了粉防已碱对血管平滑肌细胞增殖的作用及对原癌基因和抑癌基因的影响。结果发现：粉防己碱能逆转血管紧张素Ⅱ所致的H3-胸腺嘧啶核苷掺入量增多，阻止血管平滑肌细胞由静止期进入DNA合成期和有丝分裂期，能使c－fos、c—myc、c－sis原癌基因相关抗原及mRNA表达减弱，使P53抑癌基因相关抗原及mRNA表达增强。本文提示：粉防己碱有抑制血管平滑肌细胞增殖作用，同时能影响癌基因表达。

In order to elucidate the effects of tetrandrine on angiotensin Ⅱ-induced vascular smooth muscle cell proliferation and expression of oncogenes c-fos, c-myc, c-sis and antioncogene P53, the experimental models of proliferation of cultured porcine aortic smooth muscle cells induced by angiotensin Ⅱ were established, and H3-thymidine incorporation, flow cytometry, immunohistochemistry and Northern blot were used. The results showed that tetrandrine could drop H3-thymidine increased by angiotensin Ⅱ and holdback vascular smooth muscle cells from static phase (G0/G1) to DNA synthetic phase (S) and mitotic phase (G2/M). Furthermore, tetrandrine could reverse the enhanced expression of oncogene c-fos, cmyc, c-sis and reduced expression of antioncogene P53 induced by angiotensin Ⅱ. These results suggest that tetrandrine may inhibit DNA synthesis and vascular smooth muscle cell proliferation, and is related with the mechanism of molecular biology of controlling oncogene.
