硬组织替换材料骨界面细胞PKCα介导的信号传递实验研究

摘要 为阐明骨组织替代材料影响细胞代谢机理,从信号传递角度考察蛋白激酶C,在材料与细胞作用中的地位,本实验将四种骨组织替换材料分别植入SD大鼠胫骨人工缺损处,分3、5、7、9、11天五个阶段观察,运用定量免疫组织化学的方法,考察材料对界面细胞PKCα表达的影响。结果显示:PKCα介导的信号传递在材料与细胞作用中只是一种辅助途径。 Objective: To elucidate the mechanism of the interactiion between biomaterials and cells by investigatingthe effect of PKC expression of bone- derived cell on the interaction betWeen biomaterials and cells .Method:Four kinds of hard tissues substitute materials were respectively implanted in the tibia defects of 3-month-Oldrats. The defect was under the epiphyseal disk 1-2mm with diameter 2mm.The specimens from control andexperimental groups were examined at 3, 5, 7, 9, 11days after operation. It was studied the effect of biomaterialon the expression PKCa of bine-derived cell by immunohistochemical technique. Results: the biometerialcould affect the expression of PKCa, but there were some differences between the change of PKCa and that ofthe morphological Observation(ossification and reconstruction). Conclusion: PKCa only play an auxillary rolein the interaction between material and cell.
